Abstract

Streptomyces merupakan aktinomiset tanah yang berpotensi sebagai mikroba pemacu pertumbuhan tanaman dan agen biokontrol. Namun, keberhasilan aplikasinya di lapangan sering tidak konsisten akibat tekanan lingkungan dan rendahnya stabilitas formulasi. Penelitian ini bertujuan menganalisis kemampuan matriks natrium alginat dan kalsium klorida dalam mempertahankan viabilitas inokulum Streptomyces enissocaesilis strain RFZm-Pg. Selain itu, penelitian ini mengevaluasi karakteristik fisik enkapsul melalui pengamatan morfologi SEM, pembengkakan, kehilangan massa, dan viabilitas inokulum terenkapsulasi di tanah steril. Inokulum diperoleh dari medium beras-jagung, kemudian
disuspensikan dalam natrium alginat 2% (b/v) dan dienkapsulasi melalui gelasi ionik menggunakan CaCl₂ 0,1 M. Viabilitas inokulum dalam larutan alginat dan setelah enkapsulasi dianalisis menggunakan metode hitung cawan. Pengamatan pembengkakan dan kehilangan massa dilakukan pada tanah steril menggunakan kantong nilon. Hasil menunjukkan bahwa viabilitas inokulum dalam larutan alginat meningkat dari 1,3 × 10^7 CFU/mL pada menit ke-0 menjadi 4,5 × 10^7 CFU/mL pada menit ke-30, kemudian menurun menjadi 4,5 × 10^6 CFU/mL pada menit ke-180. Viabilitas pasca-enkapsulasi mencapai 2,5 × 10^8 CFU/g. Enkapsul menunjukkan pembengkakan awal sebesar 11,9%
pada hari ke-3 dan kehilangan massa kumulatif sebesar 70,3% setelah empat minggu. Viabilitas inokulum terenkapsulasi di tanah meningkat dari 4,7 × 10^3 menjadi 7,6 × 10^5 CFU/g selama empat minggu. Hasil ini menunjukkan bahwa matriks natrium alginat-kalsium klorida mampu mempertahankan viabilitas inokulum dan mendukung pelepasan bertahap Streptomyces enissocaesilis strain RFZm-Pg di tanah steril.
